We prepare and isolate recombinant proteins and also make mutants to investigate their mechanism of function, exploring spectroscopic, electrochemical and chromatographic techniques.<a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S0065291117300218?via%3Dihub\"><img decoding=\"async\" loading=\"lazy\" class=\"aligncenter size-large wp-image-437\" src=\"https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/heme-based-1024x724.jpg\" alt=\"\" width=\"640\" height=\"453\" srcset=\"https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/heme-based-1024x724.jpg 1024w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/heme-based-300x212.jpg 300w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/heme-based-768x543.jpg 768w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/heme-based-1536x1086.jpg 1536w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/heme-based-2048x1448.jpg 2048w\" sizes=\"(max-width: 640px) 100vw, 640px\" \/><\/a><br \/>\nFixL has been a prototype of heme-based sensor kinase proteins and much have been learned about it. One interesting correlation of midpoint eletrochemical potential of the heme with kinase activity was proposed by our lab during studies with a hybrid FixL from <em>R. etli<\/em>.<br \/>\nOur independent work has showed DevS from <em>M. tuberculosis<\/em> has a peculiar mechanism of signal transduction, which upon binding to oxygen the quaternary structure is mainly dimeric and tetrameric maintaining histidine kinase activity off. This changes under anaerobic conditions when octameric structures are quickly formed allowing histidine kinase to become active.<br \/>\n<a href=\"https:\/\/febs.onlinelibrary.wiley.com\/doi\/10.1111\/febs.14734\"><img decoding=\"async\" loading=\"lazy\" class=\"aligncenter wp-image-436 size-large\" src=\"https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/DevS_Josi-1024x724.png\" alt=\"\" width=\"640\" height=\"453\" srcset=\"https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/DevS_Josi-1024x724.png 1024w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/DevS_Josi-300x212.png 300w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/DevS_Josi-768x543.png 768w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/DevS_Josi-1536x1085.png 1536w, https:\/\/bioinorganica.ufc.br\/wp-content\/uploads\/DevS_Josi-2048x1447.png 2048w\" sizes=\"(max-width: 640px) 100vw, 640px\" \/><\/a>In another work, a detailed spectroelectrochemical study of DevS showed its reasonably high potential in agreement to its role as an oxygen sensor, discarding a redox sensing activity.
